XCapSeq™ TMB 680 Gene Probe Combination

Product Introduction      
           The XCapSeq™ TMB 680 Panel consists of over 19,000 XCapSeq probes, covering 680 genes and their related loci. The probe region covers 2.25M of the human genome and can comprehensively assess immune, targeted, and chemotherapy-related loci in tumor detection. The concentration of each probe is known and can be added as needed.

           High Clinical Value Product Design

All testing modules have undergone rigorous design. The solid tumor module covers approximately 2.25M, encompassing 680+ genes, including the entire exonic regions of 573 genes related to the occurrence, development, and precision treatment of solid tumors. It also integrates two major internationally recognized panels: FoundationOne™ and MSK-IMPACK™ gene sets.  
           High-Performance DNA Probe Panel  
           Based on a proprietary single-strand independent DNA synthesis method, each part of the panel can be deeply optimized, resulting in superior performance. High-depth sequencing of the 2.25M solid tumor panel can achieve >99.5% coverage of target sequences at >500×.  
           Precise Low-Frequency Detection Capability  
           Based on UMI molecular markers and two-round error correction and denoising algorithms, the lower limit of mutation frequency detection is as low as 1/1000, significantly reducing the false positive rate.  
 

 

Figure 1. The XCap TMB 680 Panel demonstrates excellent capture efficiency and uniformity. Sequencing a 300bp DNA library with a hybridization time of 2 hours and three replicates per sample shows that at 0.2x average depth, uniformity can reach 99.8%, and at 0.5x average depth, uniformity can reach over 90%, with capture efficiency (reads) greater than 80% and (base) greater than 70%.

 

 

Table 1. The XCap TMB 680 Panel demonstrates excellent accuracy. Using the horizon 0.1% cfDNA standard product, the XCapSeq TMB 680 Panel obtained similar mutation frequencies. This indicates that the XCap TMB 680 Panel is fully suitable for ctDNA detection.
