The most commonly used methods for processing flow cytometry tissue specimens are enzymatic digestion and mechanical methods.

• Enzymatic digestion is time-consuming, and different tissues require different enzymes. Surface antigens are easily affected after digestion, so its application is limited. Therefore, most people still try to use mechanical methods.
• Many commercialized mechanical grinding machines are now available, such as those from BD and Miltenyi, but they are expensive.

Which is better, these grinding machines or manual methods?

The authors studied diseases such as diffuse large B-cell lymphoma, comparing 51 machine-ground, 27 manually-ground specimens, and 15 specimens combining machine and manual grinding of tissues.

Manual method: Tumor clone detection rate was 88.9%

Combination of machine and manual: Detection rate was 66.7%

Machine grinding: Only 62.7%
