Currently, the most common methods for processing flow-organized specimens are enzymatic digestion and mechanical methods.
• Enzymatic digestion is time-consuming, and different tissues require different enzymes. The surface antigens of cells are easily affected after digestion, thus limiting its application. Therefore, the mechanical method is still mostly preferred.
• Many commercialized machines for mechanical grinding are now available, such as those from BD and Miltenyi Biotec, but they are expensive.
Which is better, these grinding machines or manual methods?
• The author studied diseases such as diffuse large B-cell lymphoma, comparing specimens from 51 cases of machine grinding, 27 cases of manual grinding, and 15 cases combining machine and manual grinding.
• Manual method: Tumor clone detection rate was 88.9%
• Combined machine and manual method: Detection rate was 66.7%
• Machine grinding: Only 62.7%
