Product Introduction  
           The XLIBPreP™ One-Step Enzymatic Library Preparation Kit is a universal, rapid, and efficient sequencing library construction kit compatible with Illumina and MGI high-throughput sequencing platforms. This product uses an enzymatic method to fragment large DNA fragments, simultaneously performing end repair and A-tailing. No purification is needed between steps; the product can proceed directly to sequencing adapter ligation. This product is suitable for various sample types, including genomic DNA (gDNA), formalin-fixed paraffin-embedded DNA (FFPE DNA), and chromatin immunoprecipitation DNA (ChIP DNA), and exhibits good compatibility with different DNA input amounts. This product offers a simple and rapid library construction workflow, enabling completion of library construction experiments within 2 hours, significantly reducing operation time.

Note: For library construction using 200ng of gDNA, with different enzymatic digestion times for two sample replicates, the XLIBPrep Enzymatics Library Kit demonstrates stable performance and high yield, with a single band. As the enzymatic digestion time increases, the length of the digested fragments decreases. By adjusting the enzymatic digestion time, different digestion lengths can be expected. Only 5-10 minutes of enzymatic digestion time is required to obtain a library size of 300-500 bp.

Note: Sample grading standard | FFPE A: >2 kb with bright band and no trailing bands; FFPE B: > 2 kb with faintly visible main band, main band below 2K, exhibiting moderate smearing; FFPE C: Bands mainly distributed at 200-500 bp, smear-like. The XLIBPrep Enzymatics DNA Library Kit shows consistent and single band distribution for library construction from DNA with different degrees of degradation.

Note: After library construction of the LDT –C91 (purchased from LDT Biosciencs) standard, followed by capture sequencing using the XCapSeq 120 Hotspot Panel and XCapSeq Hyb&Wash Buffer Kit, and then sequenced using NovaSeq 6000, it can be seen that XLIBPrep Enzymatics DNA Library Kit shows the best capture efficiency and the best uniformity in the captured data.
