Product Introduction  
           XLIBPreP™ Fast Library Prep Kit is a universal, rapid, and efficient sequencing library construction kit compatible with Illumina and MGI high-throughput sequencing platforms. This product repairs and adds A-tails to DNA that has been chemically, enzymatically, and physically fragmented, eliminating the need for purification steps before proceeding directly to the sequencing adapter ligation experiment. Through optimization of the ligation module system, the DNA ligation efficiency is maximized, regardless of DNA end integrity, DNA starting amount, or adapter concentration. This product exhibits good compatibility with DNA starting amounts from 1 to 500 ng and is suitable for various sample types, including genomic DNA, FFPE DNA, cell-free DNA (cfDNA), and ChIP DNA. It demonstrates superior performance, particularly in library construction quality for FFPE and cfDNA samples with low DNA starting amounts. This product provides a simple and rapid library construction workflow, enabling completion of the library construction experiment within 2.5 hours, significantly reducing the operation time.

Note: Library construction was performed on LDT –C91 (purchased from LDT Biosciencs) standards after mechanical fragmentation using bioruptor pico. All input amounts represent three replicates of library construction. XLIBPrep Fast Library Kit showed stable performance, high yield, and a single band. WGIA represents XLIBPrep Fast Library Kit, while Ventor A and Ventor B are competitors.

Note: Sample grading | FFPE A: ~2kb with a bright, single, non-trailing band; FFPE B: 500~2000 bp moderate dispersion; FFPE C: bands are mainly distributed between 200-500 bp and are diffuse.

Note: After library construction on LDT –C91 (purchased from LDT Biosciencs) standards, re-sequencing was performed using NovaSeq 6000. Mapping rate, duplication rate, and SoftClip Rate were statistically analyzed. XLIBPrep Fast Library Kit has lower duplication values and lower rates of Softclip Reads and Chimera.

Note: After library construction on LDT –C91 (purchased from LDT Biosciencs) standards, target capture sequencing was performed using XCapSeq 120 Hotspot Panel and XCapSeq Hyb&Wash Buffer Kit, followed by sequencing on NovaSeq 6000. XLIBPrep Fast Library Kit showed the best capture efficiency and the uniformity of the captured data meets the needs of clinical tumor detection.
